Short Description

8 B-cell hybridomas and derivative monoclonal antibodies

Abstract

These murine mAb specificity for ferret immunoglobulin (Ig). These mAb have a variety of documented research, and potentially veterinary, applications including flow cytometry (FCM), enzyme-linked immunosorbent assay (ELISA), enzyme-linked immunospot (ELISPOT) and Western blot (WB). These reagents address a currently unmet need because they enable discrimination of ferret B cells and derivative products (Ig) using mAb, whereas existing reagents are polyclonal in nature and finite in quantity. These mAb also enable segregation of ferret B cells by FCM on the basis of light chain expression, and thus are invaluable in diagnosis of B cell malignancies. Moreover, these mAb are amendable to intracellular staining and enable identification/enumeration of antibody-secreting cells (ASC) by FCM. These reagents also have application in ELISA and can be used as either coating antigen or secondary detection reagents. When used in combination as coating antigen in an ELISA, these reagents would enable quantification of total ferret Ig. If a single mAb is used as coating antigen in an ELISA, the reagent enables quantification of total kappa or lambda expressing ferret Ig. If used as a secondary detection reagent, these mAbs enable perception of either total antigen-specific ferret Ig or distinction between antigen-specific kappa or lambda encoding ferret Ig. Similar to their applications in ELISA, these same mAbs can be used in the ELISPOT assay to enumerate either total or antigen-specific ASC. Finally, mAbs have applications in the WB assay, and enable detection of free Ig light chains (indicator of B cell malignancies such as multiple myeloma).

Mouse Monoclonal Antibodies Mouse anti-Ferret Igk

  • GK1-1-4-B10 (IgG1,k) – this mAb has application in ELISA, ELISPOT, Flow Cytometry and Western Blot. This mAb is also useful for intracellular staining and identification of Ferret antibody-secreting cells (ELISPOT + FCM).
  • GK1-1-4-E3 (IgG1,k) - this mAb has application in ELISA, ELISPOT and Flow Cytometry.

 Mouse anti-Ferret Igl

  • GK1-1-4-D11 (IgG1,k) - this mAb has application in ELISA, ELISPOT and Flow Cytometry.
  • GK1-6-H5 (IgG1,k) - this mAb has application in ELISA, ELISPOT and Flow Cytometry.
  • GK1-2-8-H9 (IgG1,k) - this mAb has application in ELISA, ELISPOT, Flow Cytometry and Western Blot. Existing data suggests that this mAb recognizes an epitope shared by all non-Ig+ expressing Ferret B cells. This mAb is also useful for intracellular staining and identification of Ferret antibody-secreting cells (ELISPOT + FCM).

Mouse anti-Ferret IgH

  •  GK1-2-6-B5 (IgG1,k) - this mAb has application in Flow Cytometry. Existing data suggests that this mAb recognizes all Ferret B cells by FCM, although conjugation with NHS-ester (labeling free amines) destroyed antigen specificity (may need to chemical conjugate using alternative chemistry).
  • GK1-1-6-B7 (IgG1,l) - this mAb has application in ELISA, ELISPOT and Flow Cytometry. Existing data suggests that this mAb recognizes all Ferret B cells by FCM.
  • GK1-2-11-E3 (IgG1,k) - this mAb has application in ELISA, ELISPOT, Flow Cytometry and Western Blot. Existing data suggests that this mAb recognizes an epitope that is restricted to secreted immunoglobulin, and present on the Ig chain. This mAb is also useful for intracellular staining and identification of Ferret antibody-secreting cells (ELISPOT + FCM).
